Nitrogen isotopic composition of microbial biomass grown in the laboratory under N2-fixing conditions
Nitrogen isotopic composition of microbial biomass grown in the laboratory under N2-fixing conditions. Also included are meta-data that describe the experimental conditions. The conditions were chosen to mimic specific parameters of the early Earth, in particular variable CO2 and O2 supplies, to test if d15N data recorded in ancient biomass in the rock record is affected. Also included are data from the wider literature for comparison, as well as the results of statistical tests.

Details about the culturing experiments and isotopic analyses are contained in a manuscript by Wannicke et al. (in press) "Exploring the influence of atmospheric CO2 and O2 levels on the utility of nitrogen isotopes as proxy for biological N2 fixation" Applied and Environmental Microbiology. In brief, cultures were set up in flasks within gas-controlled chambers. The biomass was collected after several days, centrifuged and freeze-dried. The dry biomass pellets were analysed for nitrogen isotopes by flash combustion in an Elemental Analyzer coupled to a gas-source mass spectrometer (EA Isolink, ConfloIV, MAT253 IRMS).
